The intention of the study was to test the hypothesis that tryptophan concentrations in plasma are a function of subjective response to ingestion of either an amino-acid or a carbohydrate. Twenty-four patients (18 normal subjects and 15 symptomatic), with a mean age of 26 ± 13 years (range 24-27 years), were randomly allocated to an intake of either 300- and 400-mg tryptophan and carbohydrate, or 800- and 1500- mg tryptophan and carbohydrate. The patients were assessed weekly on the Beck Depression Inventory, the Montgomery-Asberg Depression Rating Scale (MADRS), the Hamilton Rating Scale for Depression (HAM-D), the Hamilton Anxiety Symptom Scale (HAM-A), and the Self-Reflection Inventory. As a measure of subjective response to dietary supplements, subjects’ ratings of hunger and the urge to eat (mictroracial and somatic) were assessed using a food item response technique. There was a significantly higher (P < 0.001) need to eat for the placebo (600-mg tryptophan), 800-mg tryptophan, and 1500-mg tryptophan groups vs. the control group (p < 0.001).
